The HPE FlatTop Tower and the HPE gels have been developed together as a system, in order to achieve better results than can be obtained with conventional SDS polyacrylamide gel electrophoresis (PAGE) technology.

The HPE Tower System
The SERVA HPE tower consists of four horizontal electrophoresis chambers, which are built as movable drawers into a metal housing. The flatbed gels are placed on aluminium oxide ceramics cooling plates, which ensures very efficient heat dissipation and therefore straight electrophoresis migration in each gel. The HPE tower also has a built-in pump for optimum cooling water flow through the plates. When needed, electrophoresis buffer is supplied by fibre wicks, soaked with concentrated buffer. The electrode positions are adjustable to different gel sizes. The HPE tower runs with an external power supply (HPE-PSP) and a thermostatic circulator (cat. No. HPE-CU1), both included with the HPE tower system (HPE-TS1).

To complete the HPE technology flatbed gels for different applications are available:
2D SDS PAGE Gel Kits
2D SDS Page gel kits are available in three different formats:
• HPE Large Format Gels: suitable for running 1 x 24 cm IPG Strip + marker lane
• HPE Double Gel Kits: suitable for running 2 x 11 cm IPG Strip + marker lane
• HPE Triple Gel Kits: suitable for running 3 x 7 cm IPG Strip + marker lane
All gels can be run by horizontal electrophoresis on HPE Tower and BH-2C. Gels are on standard backing for Silver or Coomassie staining or on non-fluorescence (NF) backing for best results with fluorescent staining and labeling. Each kit contains 4 gels + buffer kit (including running and equilibration buffer, buffer wicks and cooling contact fluid).

1D SDS PAGE Gel Kits
There are different gel types and application optimized buffer kits available:
| • | 1D SDS PAGE gel kits are ready to use kits for SDS PAGE. The gel concentration of the gels are 10, 12.5 or 15 % (T) acrylamide. They are available with 25 (type 25S for 15 µl/slot) or with 52 (type 52S with 6 µl/slot) slots. Gels are on standard backing. Each kit contains 4 gels + buffer kit. All gels can be run by horizontal electrophoresis on HPE Tower and BH-2C. |
| • | CleanGels are rehydratable gels are available with acrylamide content of 10 or 15 % (T) and with 25 (type 25S for 15 µl/slot) or with 52 (type 52S with 6 µl/slot) slots. Gels are on standard backing. A special formulated ampholyte mixture for CSF analysis on CleanGels is available. |
| • | Sortex-Kit for Cereal-cultivar-differentiation-electrophoresis. Contains 4 gels on standard backing and special buffer kit for flatbed electrophoresis on HPE tower and BH-2C. |
| • | Native Buffer Kit contains buffer system for native cathodal flatbed electrophoresis of proteins on HPE tower and BH-2C. |

The Precast 2D Vertical Gel Kits
The 2D DALT gel kits contain 6 or 12 gels for ETTAN DALTsix and ETTAN DALTtwelve, respectively. They contain 6 or 12 gels + buffer kits containing anodal and cathodal buffers as well as IPG equilibration buffer and agarose for strip fixation.

Flatbed Gels for Isoelectric Focusing (IEF)
The FocusGels are rehydratable gels for flatbed isoelectric focusing on HPE Tower and BH-2C. They are available in 9 different pH ranges. The gels have none, 24 (type 24S with 25 µl/slot) or 40 (type 40S with 12 µl/slot) slots. Ampholyte mixture for rehydration and dilution buffer required for running IEF gels is available.

SERVASnow Staining Kit
Rapid, sensitive and completely reversible protein staining method

The SERVASnow Staining Kit is a zinc-based protein gel stain as sensitive as other commonly used protein staining techniques, but with the great advantage of being erasable. The simple staining protocol results in a stained gel with an opaque milky-white background and clear protein bands. The white background is easily removed by incubation in a mobilizing buffer. The gel can then be restained by any other method or the proteins can be transferred to a membrane for Western Blotting or eluted from an excised gel piece.bands. Agarose SERVA Wide Range (molecular biology grade) is ideal for rapid separation of nucleic acids from 250 bp to 23,000 bp. SERVA DNA Stain G is a safer alternative to traditional ethidium bromide stain for detecting nucleic acid in agarose gels. It is at least as sensitive as ethidium bromide and can be used in exactly the same way in agarose gel electrophoresis. SERVA DNA Stain G can be added to the gel solution, working dilution is 1:20.000 to 1:50.000.

SERVA ICPL™ Quadruplex Kit

The powerful ICPL™-technology for comparative quantification of proteins is now available with a fourth label. Applying the ICPL™ Quadruplex method the simultaneous quantitative analysis of four independent proteome samples can be performed by stable isotope protein labelling of all lysine residues and protein N-termini. The ICPL™-technology combines the power of stable isotope coded labelling with the concept of reducing the proteome complexity by fractionation of intact proteins.
Data evaluation with ICPLQuant – freeware and easy to use
A new software - ICPLQuant – was developed for analysis of LC/MS data of ICPL™ – labelled proteins (Brunner et. al., ICPLQuant – A software for non-isobaric isotope labelling proteomics, Proteomics 10, 2, (2010)). The software tool is free of charge and can be downloaded from here.
The main functionalities are peptide pattern searching (ICPL duplex, triplex, quadruplex), peptide quantification in an LC-run and merging with MASCOT protein results. ICPLQuant takes deisotoped data as input, generates precursor files for MS/MS and is able to import MASCOT result files for combining MS/MS and quantification results.
By selecting only regulated precursors for subsequent MS/MS experiments, the software minimizes time consuming acquisition and interpretation of MS/MS data. Up to now, ICPLQuant is able to handle data generated by MALDI (Applied Biosystems 4700/4800, Bruker Ultraflex) and ESI (Thermo Orbitrap) machines.

Reliable, efficient and easy sample preparation is a crucial step for achieving optimal and reproducible results in proteomics. The SERVA BluePrep Kits cover sample preparation steps from protein isolation, concentration, buffer exchange, removal of interfering substances, tryptic digestion of proteins to abundant serum protein depletion or recombinant protein extraction from E. coli.
