DNA Polymerase I Klenow Fragment, 3 U/µl

Solution in in 100 mM Tris-HCl (pH 7.5), 10 mM 2-mercaptoethanol and 50 % glycerol. RNases, Nickases, Exo- and Endonucleases not detected.
Supplied with 10 x Assay Buffer: 500 mM Tris-HCl (pH 7.6 at 25 °C), 100 mM MgCl2.
The large fragment of DNA polymerase I, Klenow Fragment, has two of the three properties of the intact DNA polymerase I enzyme. Klenow Fragment has 5’ → 3’ DNA polymerase activity and 3’ → 5’ exonuclease activity. However it lacks the 5’ → 3’ exonuclease activity of intact DNA polymerase I.

Unit Definition: One unit is defined as the amount of enzyme required to convert 10 nmoles of total deoxyribonucleotides to an acid insoluble form in 30 minutes at 37 °C.
